Revisiting mRNA tailing by 3′-terminome sequencing

Abstract:

The 3′ termini of eukaryotic RNAs reflect the history of transcript and play important roles in determining the fate of RNA. Despite the importance, the actual sequences of 3′ ends remain unknown for the vast majority of transcripts. Genome scale investigation has been hampered due to the limitation of the sequencing technologies which cannot determine homopolymeric sequences of longer than ~30 nt. We developed a new method, TAIL-seq, to sequence the very end of mRNA molecules using alternative signal processing procedure. For the first time, TAIL-seq allowed us to extract detailed transcriptome-wide information on sequences at 3′ ends. TAIL-seq has discovered a new type of vertebrate poly(A) tail modification, U tailing. The additional U tails often follow after short poly(A) tails. They are found to be essential for inherent kinetics in catabolism of most messenger RNAs. The U tailing as a decay signal is also conserved among mRNAs of several metazoan species. We report that two terminal uridylyltransferases,TUT4 and TUT7, have crucial roles in decay of maternal transcripts in the maternal-to-zygotic transition of vertebrate embryos by uridylation of mRNAs.
